These findings demonstrate that glycolysis is dispensable for macrophage effector function in tissue-resident alveolar macrophages and highlight the importance of macrophage tissue origin (tissue resident vs. recruited) in immunometabolism. FEBRUARY 2020 AJRCMB Rats with a Human Mutation of NFU1 Develop Pulmonary Hypertension This study describes the first humanized genetic rat model to spontaneously develop pulmonary arterial hypertension (PAH). Rats with deficient NFU1 produced a strong PAH phenotype with proliferative changes in the lung vasculature and right-heart hypertrophy. The authors also provide a molecular characterization of the model. FEBRUARY 2020 AJRCMB Alveolar Stretch Activation of Endothelial Piezo1 Protects Adherens Junctions and Lung Vascular Barrier These data demonstrate that downregulation of Piezo1 signaling in endothelium is a critical factor in the pathogenesis of ventilator-induced lung injury, and thus augmenting Piezo1 expression or pharmacologically activating Piezo1 signaling may be an effective therapeutic strategy. The convincing recent clinical trial data on the lack of significant toxicity in high-risk populations justify the timely conduct of clinical studies to achieve “repurposing” or “repositioning” of pioglitazone for the treatment of clinical PAH.
